Description
CJC-1295 without DAC — also called Modified GRF(1-29) or Mod GRF 1-29 — is a more stable synthetic version of GHRH(1-29), the 29-amino acid fragment of GHRH that is sufficient to fully activate the GHRH receptor. Four targeted amino acid changes improve its resistance to the blood enzyme that rapidly breaks down natural GHRH, extending its active window from around 7 minutes to around 30 minutes. Unlike the DAC variant, it circulates freely and produces discrete GH pulses more analogous to the body’s natural GH secretion rhythm.
What the research has found
Studies confirmed the four amino acid substitutions collectively extend plasma half-life 4–5 fold over native GHRH, providing a substantially more practical research tool without requiring albumin conjugation (Bowers, 1998). When combined with ipamorelin in rodent models, CJC-1295 without DAC produced synergistic GH release significantly greater than either compound alone — establishing this pairing as a standard research approach for maximal GH axis stimulation (Bowers, 1998). Comparison studies with the DAC variant confirmed the non-DAC form produces distinct GH pulses with normal inter-pulse troughs, making it the preferred tool when research requires studying physiological pulsatile GH dynamics (Teichman et al., 2006). Research also established that CJC-1295 without DAC preserves the same receptor binding activity and pituitary signalling as native GHRH, making it a direct replacement in experimental protocols where metabolic stability is required (Bowers, 1998).
