Description
KPV is a tiny three-amino-acid fragment derived from alpha-MSH, one of the body’s natural anti-inflammatory signals. What makes it interesting to researchers is that it appears to carry the anti-inflammatory properties of its parent molecule — without any of the tanning or hormonal effects. Most research has focused on its behaviour in gut and skin tissue models.
What the research has found
• Studies in intestinal cell models showed KPV significantly reduced the release of inflammatory signalling molecules — including IL-8 and TNF-α — when cells were exposed to inflammatory stimuli. Effects were seen at very low (nanomolar) concentrations, suggesting high potency relative to dose (Dalmasso et al., 2008)
• A key discovery was that KPV is actively transported inside intestinal cells via a specific transporter called PepT1, allowing it to act directly on inflammatory pathways from within the cell. This is unusual for a peptide and may explain why it appears particularly relevant to gut inflammation research (Dalmasso et al., 2008)
• In mouse models of colitis, KPV administered directly to the intestinal lining reduced visible inflammation markers, lowered inflammatory enzyme activity, and decreased cytokine levels compared to untreated controls (Kannengiesser et al., 2008)
• Research has also identified effects on multiple inflammatory signalling pathways — including NF-κB and MAPK — suggesting KPV may suppress inflammation through more than one mechanism simultaneously (Brzoska et al., 2008)
